N-hydroxy succinyl acetate coating crosslinking agent:The NHS ester can form very stable amide bonds under the physiological pH condition and in the form of a beramine-amino acid. The protein n-amino and lysine residue of n-amino and lysine residue can form amide bonds with NHS ester.
The NHS ester, which forms a very stable amide bond, releases n-hydroxysuccinamide. The water in the NHS is hydrolyzed, and the rate of hydrolysis increases with pH.
If the concentration of protein is low, then the NHS's hydrolysis reaction will be stable. The NHS ester can have a specific crosslinking reaction with the albumin in a non-beramine-based ion buffer solution. A common buffer system includes a phosphate buffer between 50 and 20mmol/L, a carbonate buffer, a HEPES or a boric acid salt buffer solution.
Tris is not found in the solution of the NHS ester's reaction, but it can be used for the suspension of the NHS esters. Normally, the treatment of NHS esters in neutral and alkaline conditions can be stopped by adding an excess of Tris at the end of the reaction.
